In vitro, IL6 is constitutively created by Sertoli cells and Leydig cells.375,386,390,50204 Production of IL6 in rat or mouse Sertoli cells is stimulated by a selection of agents, which includes FSH, testosterone, IL1, TNF, and LPS, and is inhibited by INFG.387,496 Phagocytosis of residual cytoplasm shed by late spermatids in the time of spermiation triggers Sertoli cell IL1 production, which stimulates IL6 secretion in an autocrine manner, by activating leukotriene production by means of the lipoxygenase pathway (Figure 19.10).375,387 Leydig cells are robust producers of IL6 soon after stimulation by LH, LPS, or IL1 in vitro, and may well be the important supply of this cytokine within the testis.390,503,504 Humanperitubular cells express IL6 mRNA, but possibly not the protein, when stimulated by TNF,478 and isolated spermatogenic cells from adult mice express IL6 mRNA and protein when stimulated using a TLR3 ligand, poly(I:C).451 Although not as intensively studied within the testis as either IL1 or TNF, it seems that IL6 regulates the function of somatic, spermatogenic, and immune cells.
